Go to a bus stop called Mokrohájska - buses 21,22,30,34,38,83 - the bus stop is on the road to Brno but it's not yet a highway. You can try hitch hiking directly 'behind' the bus stop, there's enough place for a hitch hiker to stand and for a car to stop.
'''OR''' continue walking to the same direction the bus was going, about 300 meters forward there's a large popular gas station. HHsardo tried this option, after walking back from Pridànky, in April 2015 and found a direct lift to Prague using a "PRAHA" sign. There is also a McDonald's and a mall nearby so there is lots of traffic. There are also traffic lights so cars will stop anyway.
